Nezuko experiences a harrowing transformation into a demon following the tragic slaughter of her family by demons. Unlike many of her kind, she maintains her human emotions and a deep sense of empathy. Even with her demonic traits, Nezuko is fiercely protective of her companions. Furthermore, she is characterized by her quiet nature, communicating mainly through her actions instead of verbal communication. Shy and clingy, she is unable to speak because her mouth is covered by a bamboo muzzle to prevent accidental biting. Nezuko has the ability to shapeshift into both younger and older versions of herself.

As the younger sister of Tanjiro Kamado, Nezuko is one of the last two survivors of the Kamado family. She was once human but was attacked and transformed into a demon by Muzan Kibutsuji.

Nezuko is depicted as a young girl with pale skin, large, prominent fangs, a slender figure, and sharp nails that are light pink at the base and fade to a red-pinkish hue at the tips. Her long, wavy black hair extends just below her waist, transitioning to a flame-orange color past her elbows, styled with crimped large ridges and swept to the left. She has soft, pale pink eyes that appear lighter around the edges of the irises and display a Persian pink shade beneath the pupils; they slope down towards the sides of her face, accentuated by long eyelashes. When she transforms, her pupils turn into black slits. According to Tanjiro, Nezuko was renowned for her beauty in their hometown.

Her attire consists of a light pink kimono adorned with an asanoha (hemp leaf) pattern, featuring a lighter pink lining layered with a white wavy yuban underneath, allowing her to move freely as a demon. The white garments cover her knees and extend down almost to her ankles, fastened with a red and white-checkered hanhaba obi, complemented by an orange-threaded obijime and a green obiage. Over this, she wears a long, dark-brown haori that reaches her calves, along with zōri sandals with pink straps and white tabi socks, wrapped in thick black materials resembling kyahan. Nezuko also has a small pink ribbon positioned on the left side of her head to keep her hair away from her face, and she wears the bamboo muzzle given to her by Giyu Tomioka, secured with a strip of red cloth around her jaw.

In her human form, Nezuko had a similar appearance, differing mainly in the absence of demonic traits, her fiery hair color, and her pale pink eyes. Instead, she had fully black hair styled in three low buns adorned with pink ribbons, and her eyes were a darker shade of pink. She wore a similar kimono without her haori and included a layered pink kimono with the white yuban beneath, complemented by a white Onbuhimo cloth used to carry her younger brother, Rokuta Kamado. After reverting to human form, these characteristics return, except for her hair, which retains its flame-orange tips—a trait passed down to her descendant, Toko.

As a human, Nezuko was a kind and nurturing individual, always considering others before herself, much like her older brother, Tanjiro. She was a responsible elder sister who prioritized her siblings’ needs. Nevertheless, she was not hesitant to put herself in danger to shield her friends from demons, even after returning to human form. Her gentle and easygoing demeanor is evident after Tanjiro apologizes to her.

Initially resembling other demons in her bloodthirstiness and hunger for human flesh, Nezuko undergoes a significant change after witnessing her brother's determination to protect her from being killed by a Demon Slayer, despite her being a demon. This leads her to become fiercely protective of him. Nezuko appears to have lost many of her human memories, retaining only those related to her family, which alters her original personality and leaves her behaving more childlike. Although she remains caring and protective towards humans she regards as family, this behavior is influenced by Sakonji Urokodaki during her two-year slumber. Nezuko still possesses some human emotions, as she has been seen to cry when upset and smile when happy, but overall, she seems more detached compared to her human peers, while still exhibiting a calmer and less malicious demeanor than most demons. She often sees reflections of her siblings in other humans, such as Rokuta Kamado in Yushiro.

Since her transformation into a demon, Nezuko has become quite bold and shows little fear in battle, fiercely safeguarding her brother and allies. Her strong willpower is evident in her refusal to consume human flesh or blood, even when faced with severe injuries or exposure to human blood, as demonstrated when she declines Sanemi Shinazugawa's attempt to entice her into biting him with his Marechi blood. This determination persists even after she is no longer a demon, as shown when she bravely jumps in front of Inosuke to protect him from her brother, who had become a demon and was trying to attack those around him. Despite the risk to her life, Nezuko does not hesitate to defend her brother.

On the rare occasions when Nezuko attempts to speak, she tends to stutter, likely due to her bamboo mouthpiece, which is rarely removed, and the fact that she has not spoken for several years following her transformation. However, after developing a resistance to sunlight, which allows her to venture outside and interact more with others, her speech has shown improvement, enabling her to pronounce words fully if they are spoken by someone else or repeated frequently enough.
