Yoko is from Littner Village, an underground weapon stronghold village, where due to a poisonous gas leak, the villagers were forced to break out to the surface and use their weapons to fight back against the Beastmen to survive.

As every member of Team Dai-Gurren, Yoko is a skilled Gunmen pilot, her Gunmen is called Yoko M Tank, a Dayakkaiser-type pink Gunmen armed with a barrel gun on its head and a cracked skull painted on it to resemble Yoko's own cracked skull hair accessory. Despite her piloting skills, Yoko mainly relies on marksmanship, only using the Yoko M Tank in situations where her weapons may not be effective against her enemies. She is an extremely skilled sniper, and her main weapon for combat is a long-range superconductivity rifle modeled after a Barrett M82 sniper rifle, guarding ammunition and a variety of weapons for emergencies {like a knife or a pistol} inside her hair, she can also use her hair chopsticks as melee weapons. Even without Spiral Power empowerment, Yoko is very fast and is surprisingly strong and proficient in martial arts for a girl of her physique.

Yoko is very confident in her sniper skills, getting flustered when people appreciate them, she also tends to get flustered when people compliment her or show her affection, at which point she becomes awkward and calls them 'idiots,' though in an embarrassed tone that differs from the 'idiot' she uses when scolding or threatening someone. Despite her no-nonsense behavior, Yoko is very cheerful and playful, loving to enjoy mundane moments with her friends, she's kinda competitive, being surprisingly invested in winning casual or playful contests, and she's blunt and sarcastic, not minding poking fun at people to hurt their ego. Boota is a pigmole with sunglasses who's cartoonishly immortal, he's Simon's pet, but he's also very attached to Yoko, as he often takes refuge between her breasts like pillows, this causes the guys to get comically jealous of the mascot, as Yoko doesn't mind Boota being there.

To fight against the Spiral King's tyranny and recover the surface for humanity, Team Dai-Gurren has risen to fight against this villains. The main form of battling in this story, both by humans and Beastmen, involves mechas, which are called Gunmen, with a large variety of Gunmen types. The main source of power in this world is called Spiral Power, present in every human being. Spiral Power is an infinite force fueled by willpower and determination that unleashes the potential of those capable of using it, being the energy used by humans to pilot Gunmen, while Beastmen use electricity. Behind the scenes, there are the Anti-Spirals, a mysterious alien race akin to humans, who believe in the Spiral Nemesis, a hypothetical cataclysm caused by the infinite evolution sparked by Spiral Power, which would eventually get out of control and lead to a universal Big Crunch. Consequently, the Anti-Spirals relinquished their physical bodies and merged their consciousness into a single extra-universal entity, the Anti-Spiral, the true antagonist of this story. This entity's objective is that once humanity reaches a certain number, it will rise to destroy Earth to put an end to the Spiral Power, just as it did with innumerous other planets that also possessed Spiral Power.
