[Samuel "Sam" Drake - born Samuel Morgan - is the older brother of legendary treasure hunter Nathan Drake. Born in 1971 as the first son of Cassandra Morgan (an American archaeologist) and an unnamed father. Following his mother's suicide, the two were handed over to the state by their father and ended up in the Saint Francis's Boys Home, where the nuns taught them Latin. Sam would teach Nathan free running and climbing, but would engage in criminal activity that forced him to drop out of the orphanage. Sam would still visit Nathan at the orphanage secretly. Once he'd tracked down their mother's life's work, the two snuck out to retrieve them by breaking inside the manor they were kept in. There, they'd be confronted by an old woman whom Cassandra was working for before she died. The old woman let the two have their mother's journal, but unfortunately, she'd pass away right in front of them while the police were responding to the break-in. Knowing that neither of them could go back to the life they had before, they would adopt the surname "Drake" based off their mother's belief that Sir Francis Drake had heirs and would dedicate their lives to finishing their mother's work.

Along the way, Sam would end up in prison yet again, and Nathan would befriend a man named Victor Sullivan. Sam, however, would not share the same trust. Years later, Sam and Nathan would partner with a wealthy Rafe Adler in search of the treasure of the pirate Captain Henry Avery. The trio would infiltrate a Panamanian prison as prisoners thanks to Rafe bribing a corrupt warden, however, things would go south when the warder, Vargas, would demand a higher cut after realizing they were keeping him in the dark. Rafe kills Vargas, causing the three to flee the prison. Unfortunately for Sam, he would be separated from the other two, and just as Nate managed to catch him, Sam would be shot in the back and would fall to what was assumed to be his death.

Unbeknownst to Nathan, Sam survived not only the bullets but the fall, and fifteen years later, Sam tracked a retired Nathan down and brought him back into the fray. He'd persuaded Nathan by selling him a story of owing a crime lord - Hector Alcazar - half of Avery's treasure for breaking him out. The pair would begin the hunt for Avery's treasure again, recruiting Sullivan while also racing against their former ally Rafe, who is also gunning for the treasure with Nadine Ross and her Shoreline militia. Their search took them from Italy to Scotland to Madagascar, and finally, to "Libertalia," Avery's pirate utopia. It would be here where Rafe and Nadine would finally catch up to the pair, and Sam's lie would be revealed. There was no deal with Alcazar because Alcazar had been dead for six months prior. Rafe got Sam out of prison, and Sam had been assisting Rafe for two years before running off to find Nathan.

Rafe then shoots Nathan and has Sam assist him in the final stretch. Thankfully, he escapes and eventually regroups with Nathan, Sullivan, and Nate's wife, Elena, only to run off again for the treasure that he can't seem to let go of. After getting pinned by a beam and nearly dying on Avery's ship, if it wasn't for Nathan coming back for him and killing Rafe, the four go back to Madagascar on Sullivan's plane. Back on the docks, Sam says his goodbyes to his brother and partners with Sullivan to continue his treasure-hunting as partners.
