Ryūko Matoi is a 17 year old girl of medium height and has blue-black hair of medium length with a single left-swept red highlight on her bangs as well as blue eyes, which have peculiar gear-like pupils with eight grooves around the edges. Her outfit at the start of the series is a white collared shirt with a red bow tie which is worn under a black & white varsity jacket with rolled-up sleeves, as well as a black skirt and white sneakers. Ryūko has a relatively simple personality: fierce and stubborn. She fears little, which is demonstrated when she directly and disrespectfully addresses Satsuki Kiryūin.

Mako Mankanshoku, is the best friend of Ryūko Matoi. Mako is a young teenage girl with brown eyes and brown hair that is styled in a bowl cut. She is often seen wearing the standard Honnōji Academy No-Star uniform; a white short-sleeve shirt with a blue neckerchief, a blue mini skirt, and a pair of white socks and light brown loafers. Mako is a hyperactive, carefree and loving girl who seems to cling to anyone she befriends, emphasized by how she becomes inseparable from Ryūko very quickly. She is also quite lazy, as shown in how she goes to sleep as soon as class begins, humorously clashing with her hyperactivity.

Satsuki is a tall, slim woman with an angular face similar to her mother's. She has thigh-length dark-blue tinted hair that has inward diamond-shaped bangs hanging over her forehead, blue eyes, fairly thick eyebrows and a large bust.She has the habit of perpetually frowning, although she does smile on rare occasions.

Honnōji Academy seemingly shared the same purpose and systems with regular school in Japan, but in actuality, it was built as the main testing ground of Life Fiber experiments (particularly Goku Uniform) commissioned by Ragyō Kiryūin herself, with Satsuki Kiryūin being charged to govern it along with the city. Because of Satsuki's totalitarian rule over both the school and the city itself, living and studying at Honnōji Academy are best described as an exercise in survival at its extreme, with its students (particularly No-Star ones) finding themselves oppressed by iron-fist disciplinarians and abusive, if not outright excessively strict club presidents. Power struggle seemed to be a recurring occurrence as promotion rewards the students better, if not outright luxurious life standards and even control over the school. Those who disobeyed the rules would be severely punished, though death sentence occurs once the offender genuinely pose a threat to the school's well-being such as attempted theft of one of Goku Uniforms.

Goku Uniforms, sometimes translated as Ultima Uniforms, are uniforms worn by high rank students at Honnōji Academy. Created by the Sewing Club, Goku Uniforms provide their wearer with incredible superhuman powers. The strength and abilities of a uniform varies by the percentage of Life Fibers it is made from, represented by the number of star-shaped markings emblazoned on them. Satsuki Kiryūin administers uniforms to students based on their skills and capabilities, which also determines their rank within the school hierarchy. One-Star uniforms are the most common variety of Goku Uniform, comprised of 10% Life Fiber. The male version is a light gray gakuran uniform with a single large four-pointed star symbol emblazoned on the center of the jacket, and a smaller white star on its collar; and the female version is a white one-piece sailor uniform with blue scarfs, collars and highlights and a small black star on the front of the collar. Two-Star uniforms are made of 20% Life Fiber and are exclusively given to club presidents (though not every club president gets one). Their appearance and application varies greatly, with the main distinction of these uniforms being the reddish lining over many surfaces, sometimes over some parts of the uniform, and always over the uniform's stars and manifested weaponry. Three-Star uniforms are the highest-ranking Goku Uniform, made from 30% Life Fiber. The Three-Star Uniforms are worn exclusively by the Student Council's Elite Four. Each uniform's design and abilities are uniquely attributed to its wearer's default capabilities.

The Elite Four are comprised of: Nonon Jakuzure, Ira gamagori, Uzu sanageyama, And Hoko inumuta.

Nonon is petite with pink hair and an angular face. Unlike most females in the series, she has a small bust and medium hips. She is almost always seen with a skull-marked hat.Although Nonon often presents a cute and sweet facade, she is a very sassy girl who possesses an extremely caustic wit. She is the most outright arrogant out of the Elite Four.

Gamagōri is a large, muscular young man who has a darker-toned complexion and short blonde hair. His eyebrows are noticeably thick and he wears a pair of small gold earrings in his enlarged earlobes, and is often dressed in his Three-Star Goku Uniform. Gamagōri is always serious, bearing no tolerance for foolishness or rule-breaking, and often shouts at the top of his lungs whenever opposed in any way. He typically berates his much more casual colleagues and keeps the students in line for Satsuki, and is considered to be her right-hand man. As befits the head of the Disciplinary Committee, Gamagōri seems to hold adherence to the law, his ideas of ethical behavior, the hierarchy of the academy and Satsuki above all else.

Uzu is a teenager with mid-length dark green hair and gray eyes. He wears a Three-star Goku Uniform, specially made into a long white coat with a high collar. It has a built-in sword sheath at the back and three spikes on each shoulder.Uzu is rather laid back and cocky, possessing absolute confidence in his abilities, always eager to test himself against worthy opponents.

Inumuta has a tealish hair color and neat hair. He has a collar-neck suit, which, at the collar, opens whenever he talks. He also wears unique eyeglasses that are tinted blue.He's highly sarcastic, often mocking others just for the sake of it and barely showing any kind of emotion other than mild amusement towards a selected few. As Inumuta is extremely protective of his data and in turn, his laptop, with any actions that can cause potential damage resulting in him losing his cool and going into a comically portrayed rage.
